- Johnny Gat and Aisha asking the Boss (after coming out of a coma and a possible gender change operation) if they did something with their hair. Julius later tries to asks the Boss the same thing during the secret mission Revelation, but is cut off them by saying, "I didn't do shit with my hair!".
- The second mission Appointed Defender is full of funny moments:
- The Boss smashes a bottle in a Brotherhood member's face. In a blink-and-you'll-miss-it moment, the text crawl on the TV news reports, "Man hit by bottle in bar".
- Johnny Gat's trial has him arguing with the judge regarding his crimes and attitude. When the Boss enters, they force a cop to drop their handgun, and it discharges. Everyone ducks for cover except Gat, and Legal Lee, Gat's lawyer, asks if anyone was shot and needs his services. The judge later comes after the Boss and Gat during gameplay with a shotgun.
Judge: Mr. Gat, you've been convicted of over 300 murders. Do you really expect this appeal to work? |
- In the closing cutscene, the Boss and Gat consider Aisha's house as an ideal gang hideout, but she sternly rejects it. They even get a little spooked by her insistence. Two hardcore gangsters are spooked by a popstar.
- The Ronin arc has many funny moments, particularly those involving Gat and Pierce:
- Pierce's intricate plan for robbing the Ronin's casino is snubbed for Gat's simple plan of shooting everyone who stands between them and the money, leaving Pierce very annoyed.
- The closing cutscene of the first mission Saints Seven, with Shogo telling Jyunichi to speak English since they're in America, and Jyunichi being jumpy. And Shogo tells Jyunichi to worry about what he tells him to rather than the casino, all while getting a blowjob from a hooker.
- In the opening cutscene for Laundry Day, Pierce gets annoyed that Jane Valderamma referred to him as an "unknown accomplice". Also, the cops in the background take a selfie with the corpse of another cop. Troy Bradshaw hired novice/incompetent cops to ensure the Saints got away with the heist, considering his time with the gang in Saints Row.
- In the opening cutscene for Road Rage, Gat brings home a Ronin member who was on Saints turf. Gat smashing his head through the glass table and then shooting him angers Aisha, due to the fact she just bought the table and that the carpet is blood-stained. Aisha is holding a mop, implying she wants Gat to clean it up.
- While driving to their airport in Orange Threat Level, Pierces changes the radio to 102.4 Klassic FM. The Boss then argues with Pierce over who has control of the radio, but the Boss wins since they're in charge and is driving the car, and changes it back to whatever they were listening to. Made even funnier if the Boss was listening to classical music in the first place.
- In Visiting Hours, the Boss escorts a bedridden Gat out of the hospital while Ronin try to kill them. The jabs they take at each other are hilarious.
- The Sons of Samedi arc has its funny moments, particularly with the Boss and Shaundi.
- In the opening cutscene for Aerial Assault, Tobias is revealed to have a pilot's license by his wife Laura, and the Boss is doubtful. Tobias enters and says, "Does it look like I'm joking?". The dialogue during the mission is also hilarious. The Boss also becomes high during the mission due to the smoke from the destroyed drug crops filling their air.

- Bad Trip as a whole, from the Boss being high in The General's limo, Mr. Sunshine's distorted appearance as a result, as well as his dialogue with Shaundi on the phone and back at the hideout.
- A great moment during the opening cutscene was with Male Voice 1 (British) Boss' exchange with Mr. Sunshine. This is made even funnier when you know that the name is in fact a slang in British English as well, and the delivery is just perfect.
- Bad Trip as a whole, from the Boss being high in The General's limo, Mr. Sunshine's distorted appearance as a result, as well as his dialogue with Shaundi on the phone and back at the hideout.

- The closing cutscene:
Shaundi: Are you sure? They'll probably send more guys to try and kill us. |
- In Bonding Experience, Pierce is oblivious to the fact the Samedi helicopters have arrived on the scene. The Boss has to angrily tell him that they've arrived. Remember, Pierce is trying to prove himself to his boss. Later, Pierce sings along to Ne-Yo's So Sick, earning the ire of the Boss.
- In Assault on Precinct 31, if the Boss has Male Voice 1 (British), he will poorly attempt to hide his accent to the police dispatcher while in the police station.
- Considering the intensity of the Brotherhood arc, funny moments are few and far between, but they still pop up.
- Carlos being annoyed at the helicopter pilot during Waste Not Want Not, when flying to the mainland from the nuclear power plant.
- While throwing fireworks during Thank You and Goodnight!, Pierce changes the radio to 102.4 Klassic FM. If the Boss has Male Voice 1, he asks Pierce, "Pierce... when did you get a vagina?". Pierce calls back to other missions where he and the Boss were fighting over the radio, saying he controls the radio since he is driving.
- In the opening cutscene for Jail Bait, one of the released Brotherhood inmates is dancing in celebration, but is hit in the crotch by a guard with their shotgun. They start dancing again, but is immediately hit in the crotch again.
- In the opening cutscene for The Enemy of My Enemy, Shaundi gets the Boss to play skee-ball with her to 'relax'. The Boss doesn't even try, with the balls rolling back down. They end up just throwing them overarm, and the balls just bounce off the machine onto the floor behind them.
- In Pyramid Scheme, it's revealed Shaundi's extensive network of ex-boyfriends finally failed to provide her with information, specifically regarding the Ultor Pyramid.
- Ultor Family Fun Day in the Corporate Warfare DLC has situations so ridiculous that it wouldn't look out of place in the third game. The boss is attacked by a gang of mimes, a Destroy UFO can be seen at one point, and a real ninja that can teleport with fancy smoke and multiply.
- Pierce and Shaundi squabbling over trying to feed the Boss information on rival gangs. One of the Male Voice 1 idle lines says Pierce should "quit bitchin' about Shaundi and just fuck 'er already".
- The Boss can be customised to be naked or look as ridiculous as possible, with a massive variety of outfits and customisation options possible. Nevertheless, they still act as serious as if they were dressed properly, and the other characters will also act the same.
- All six voices have a song they sing along to while driving that is played on 107.77 The Mix FM, and every voice also sings along to A-ha's Take On Me.
- The Boss has some funny lines while drunk or high, such as noting how Shaundi sees the world while high.
- Fuzz has several funny bits, such as pirates fighting ninjas and the cameraman's lines.
- Some of the lines during the Escort activity, such as, "Oh god, please don't be a fucking furry...".
- Adrenaline mode in Insurance Fraud causes the Boss to fly much farther when you get hit by a vehicle. It can make the Boss look like they're flying, and it makes more money.
- Several of the radio ads are funny to listen to, such as one boasting about the positives of using "NUCLEAR POWER!" with testimonials of a woman who brags about getting time off work for chemo and a man with a third arm being 50% more productive at work.

- The ad for Ship It!, where a Russian spokesperson flubs his attempts to attract people to the store, including outright threatening them and there families, much to the chagrin of the director. The tagline for Ship It! is even "Bad At Marketing, Great At Boats!"
