- James Arnold Taylor reprises his Spider-Man: Battle for New York role as Spidey himself while Josh Keaton reprises the role of Harry Osborn from the tie-in games to the first two movies, albeit he's the New Goblin here--though after The Spectacular Spider-Man, it might be odd for some people to hear them as they voiced the other's character there (with Keaton continuing on as Spidey since then).
- As an extra little bonus, in the opening cutscene Spider-Man says something that may remind fans of another work that he participated in.

- Other voices include Fred Tatasciore as Sandman, Scorpion and Carnage (for the first time); Quinton Flynn as Venom; Robin Atkin Downes as Mysterio; Roger Jackson (Mojo Jojo and the voice of Ghostface) as the Green Goblin and the Lizard; and Joe Alaskey (Daffy Duck) as Doctor Octopus.
