Mr. Tickle[]
- Catch Phrase: "I think somebody needs a TIIICKLE..."
- Friendly Tickle Torture: It's in his name. He often tries to tickle people to make them happy (whether they want him to or not).
- Innocently Insensitive: He tends to tickle people (i.e. Mr. Grumpy) even if they tell him not to, but he only tickles them because he thinks he'll make them happy that way.
- Keet: He's a cheerful, energetic guy.
- Nice Guy: Mr. Tickle is a jolly guy who wants to tickle people to cheer them up.
- Nice Hat: A blue one.
- Rubber Man: He can stretch his arms very far so that he can tickle other people.

Mr. Happy[]
- Big Fun: He's a round circle who's very cheery.
- Broken Smile: Has sported one of these a few times.

- The Pollyanna: He lives up to his name by enjoying the good things in life. Although he can be a Stepford Smiler.

Mr. Nosy & Mr. Small[]
- Affectionate Nickname: Mr. Small likes to call Mr. Nosy "Nose".
- Ambiguously Gay:
- Big Fun: Mr. Nosy is portly and mostly friendly, though he often irritates others with his nosiness.
- Drop-In Character
- Fat and Skinny: Mr. Nosy's the fat one and Mr. Small's the skinny one.
- Heterosexual Life Partners: They're very close friends who tend to be close to each other most of the time. Although the heterosexual part is a bit...ambiguous.
- Nice Hat: Mr. Small.
- Nosy Neighbor: Mr. Nosy. It's in his name, after all.
- Those Two Guys: They're by each other's side most of the time.
- Vocal Dissonance:
- In both dubs, Mr. Small has a deep voice despite his size.
- On the opposite end of the spectrum, Mr. Nosy has a rather squeaky voice in the French dub for someone as chubby as him.
Mr. Bump[]
- Butt Monkey: His defining trait is that he goes through a ton of slapstick every time he appears.
- Catch Phrase: "Poopity-poop."
- The Chew Toy: Not only is he clumsy, but he's often injured indirectly by other characters (like through Miss Whoops's obliviousness).
- Keet
- Large Ham
- Nervous Wreck: Not as much as Mr. Nervous, but he's prone to anxiety. Not that anyone can blame him, since life has it out for him.
- Oh Crap: Essentially what his Catch Phrase is.
Mr. Messy[]
- Big Ol' Unibrow: He has one, complementing his sloppiness.
- Non-Standard Character Design: Instead of having a solid shape, he's a bunch of pink squiggles.
- Odd Couple: With Mr. Persnickety/Fussy.
- The Pig Pen:
- Squick: He has some rather revolting habits, especially to a Neat Freak like Mr. Persnickety/Fussy.
Mr. Nervous[]
- Adaptation Name Change: UK dub only. In the US, his book self was always called Mr. Nervous, but in the original UK version, he was called Mr. Jelly. In this show, however, he keeps his American name.
- Blind Without'Em: Though not as much as Miss Whoops.
- Gasshole: He mentions in "Food" that radishes make him gassy. This was changed to them giving him bellyaches in the UK dub.
- Lovable Coward: He's afraid of everything, especially Mr. Tickle.
- Mr. Imagination: He often has imagination sequences about terrible things happening to him.
- Nervous Wreck: True to his name, he usually expects the worst to happen. He often runs around shrieking and panicking.
Mr. Noisy[]
- Large Ham: Given his lack of an indoor voice, he's prone to chewing scenery.
- No Indoor Voice: And it doesn't help that he often yells into a megaphone.
Mr. Lazy[]
- Adaptation Dye Job: He was white with a blue hat in the original books, but in The Mr. Men Show he is green with a red hat.
- Blinding Hat
- Brilliant but Lazy: He'd rather build machines to do the work for him.
- Lazy Bum: He often considers things to be "too much work".
- Nice Hat: He wears magenta hat that covers his eyes.
- The Stoic: He speaks in a deadpan tone and he can't emote well thanks to his hat that covers his eyes.

- Butt Monkey: Often because of Mr. Messy's messiness. He also tends to be kicked out of places for things that aren't his fault.
- Dub Name Change: In the first season, he was called "Mr. Pernickety" in the UK dub.
- Large Ham: Especially in the US dub with his pompous British-sounding accent.
- Neat Freak: As opposed to Mr. Messy, he'd rather have everything nice and neat.

- The Unintelligible: He often speaks so quietly that it's hard to understand him, moreso in the UK dub. Though in the second season of the US dub, he starts to speak more clearly.
- Vocal Evolution: In the US dub, his voice becomes clearer in the second season. This doesn't stop other characters from being unable to hear him, though.

- French Jerk: In both dubs, he has the same French accent, and as his name states, he's rude.
- Gasshole: A Running Gag is someone saying something, followed by him saying, "I'll give you (adjective)" and farting loudly. Everyone else then shouts, "Mr. Rude!!"
- Jerkass: Unsurprisingly for someone with the name Mr. Rude, he's prone to insulting others and/or farting just to annoy them.
- Running Gag: Whenever he passes gas, a bunch of Mr. Men and Little Miss angrily shout "Mr. Rude!"

- Big Ol' Unibrow: He has a unibrow that makes him look even goofier.
